Gnn contains multiple sites for posttranslational modification and a nuclear targeting sequence, and 2 Gnn isoforms contain an AAA-ATPase domain. RNase protection analysis showed high Gnn expression in mouse heart, brain, kidney, and liver, with low expression in other tissues examined. Western blot analysis detected multiple Gnn isoforms ranging from 25 to 250 kD in mouse embryos and most adult mouse organs examined. A 150-kD Gnn isoform was overexpressed in an Evi12-containing mouse leukemia cell line, but not in control cell lines.GNN expression was increased in 2 AML clusters. One cluster consisted of AML patients with a t(8;21) translocation, and the other cluster consisted of AML patients with a normal karyotype carrying an FLT internal tandem duplication.
