Using the yeast 2-hybrid system to identify potential partners of the Nesh protein, which contains an SH3 domain and proline-rich sequences similar to those of E3B1, Matsuda et al. (2001) isolated a novel full-length cDNA, which they designated TARSH. TARSH encodes a 486-amino acid protein containing an SH3 binding motif, a nuclear targeting sequence, and no catalytic domain. By study of a monochromosomal hybrid panel and radiation hybrid cell panels, Matsuda et al. (2001) mapped the TARSH gene to chromosome 3q12.SH3 domains contain about 50 amino acids that bind to proline-rich sequences and often play a role in cell growth signaling. Sequence analysis predicted that the 366-amino acid NESH protein, which is most homologous to SSH3BP1, has a central serine-rich region followed by a proline-rich region and a C-terminal SH3 domain.
