ADP Ribosylation Factor GTPase Activating Protein 2 (ARFGAP2)
Znf289 mRNA expression paralleled that of Id1 during mouse mammary gland development, with Znf289 expressed during ductal and lobuloalveolar morphogenesis when there is extensive proliferation of mammary epithelial cells. It is downregulated following the decrease in Id1 expression in differentiated, growth-arrested lactating epithelial cells.
Human ARFGAP2 lacking its N-terminal zinc finger domain bound COPI subunits, and expression of this truncated mutant inhibited COPI-dependent Golgi-to-endoplasmic reticulum transport of cholera toxin in vivo. Silencing of ARFGAP1 or a combination of ARFGAP2 and ARFGAP3 in HeLa cells did not decrease cell viability, but silencing of all 3 ARFGAPs caused cell death.
