ADP Ribosylation Factor Related Protein 1 (ARFRP1)
ARFRP1 is a membrane-associated GTP-ase and localizes to the plasma membrane. It is related to the ADP-ribosylation factor (ARF) and ARF-like (ARL) genes. The gene is located in a gene cluster that includes the a gene (M68) that is overexpressed in some tumors. The deduced 201-amino acid human ARFRP1 protein shares 97% amino acid identity with rat Arfrp1, 39% identity with human ARL3, and 33% identity with human ARF1. ARFRP1 contains all 6 of the conserved motifs presumed to mediate GTP binding. The authors found that, unlike other ARFs, ARFRP1 lacks a glycine at position 2, the site of N-myristoylation; immunoreactivity is located in the plasma membrane and not in the cytosol; and GTPase activity is expressed in the absence of tissue extracts or phospholipids.
