ARV1 encodes a deduced 271-amino acid protein that contains an N-terminal Arv1 homology domain (AHD), encompassing a zinc-binding motif and a transmembrane domain, and 5 central and C-terminal transmembrane domains. ARV1 shares 44% amino acid identity with its yeast homolog.human ARV1 complemented a yeast Arv1 mutant that displayed altered intracellular sterol distribution and was defective in sterol uptake, consistent with a role for Arv1 in trafficking sterol into the plasma membrane. The authors suggested that human ARV1 plays a role in sterol homeostasis.Under nonlethal conditions, Arv1 mutants displayed altered neutral lipid and fatty acid metabolism. Overexpressed human ARV1 suppressed the lipid metabolic defects of Arv1-null yeast cells.
