JAML is predicted to encode a type I integral transmembrane protein of 394 amino acids with a 19-amino acid signal peptide, 2 extracellular immunoglobulin-like domains, a transmembrane segment, and a cytoplasmic tail. A possible dimerization motif was also identified.
Northern blot analysis detected predominant expression of JAML in peripheral blood leukocytes, with lower expression in bone marrow, spleen, and lymph nodes as well as in fetal liver, spleen, and thymus. Western blot analysis of COS cells transfected with JAML revealed 3 bands of molecular masses 60, 58, and 50 kD, which were assumed to represent different glycoforms of the protein. In an APL cell line (NB4) treated with ATRA, only the 60-kD species was detected.
