ANAPC16 is highly conserved among vertebrates, with 95% identity between the zebrafish and human proteins. Orthologs of ANAPC6 were also found in invertebrates and plants.The deduced 110-amino acid protein has a calculated molecular mass of 11.7 kD. It has 4 putative myristoylation sites and 3 phosphorylation sites. ANAPC16 is highly conserved among mammalian species, and the human, orangutan, monkey, mouse, horse, and dog proteins are identical.Antibodies raised against ANAPC6 immunoprecipitated the entire APC/C and its cyclin B ubiquitination activity. Western blot analysis showed that ANAPC6 was present throughout the cell cycle in synchronized HeLa cells. Immunoelectron microscopy of APC/C showed that ANAPC6 localized near the CDC27 subunit at the top of the APC/C 'arc lamp' domain.
