The deduced 417-amino acid HARP protein contains 3 ankyrin repeats and a sterile alpha motif (SAM). It shares 81% and 40% overall amino acid sequence identity with the mouse homolog and the human USH1G protein, respectively.
Northern blot analysis of mouse tissues detected 3 different Harp transcripts, with predominant expression in liver and kidney; RT-PCR detected expression in liver, kidney, small intestine, colon, pancreas, and purified pancreatic cells. Immunohistochemistry localized mouse harp to epithelia of developing lung, kidney, salivary glands, and cochlea.mouse harp and harmonin in pancreatic ductal epithelial cells and a pancreatic beta cell line. HARP has a role in facilitating signal transduction in epithelia.
