Aprataxin And PNKP Like Factor (APLF)
C2ORF13 is a component of the cellular response to chromosomal DNA single- and double-strand breaks. APLF interacts with both XRCC1 and XRCC4, the same components of the single- and double-strand break repair machinery that are bound by PNK and APTX, but not with APTX or DNA ligase III (LIG3).
The FHA domain of APLF interacts with XRCC1, and that both proteins colocalize in the cell at sites of laser-induced UVA damage. The interaction of XRCC1 with APLF was largely dependent on the phosphorylation of XRCC1 by casein kinase-2 (CK2). YFP-tagged APLF accumulated in nuclear foci in the absence of XRCC1 but this was disrupted by mutations in the zinc finger-like motifs.
