Bcl2 Like Protein 14 (BCL2L14)
BCL2L14 belongs to the BCL2 protein family. BCL2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities.Three alternatively spliced transcript variants encoding two distinct isoforms have been reported for this gene.BCLGS had a more potent activity that was dependent on its BH3 domain. BCLGL with a deleted BH2 domain was as potent as BCLGS, suggesting that the BH2 domain negatively regulates BCLGL proapoptotic activity. Coimmunoprecipitation and yeast 2-hybrid analysis showed that BCLGS interacts with antiapoptotic BCL2 family members, such as BCLXL, through its BH3 domain. Confocal microscopy and cellular fractionation analysis demonstrated that BCLGS colocalizes with mitochondria independently of the BH3 domain.
