Breast Carcinoma Amplified Sequence 2 (BCAS2)
Using an 8-way (i.e., 8 cell lines) differential display analysis, followed by screening a fetal brain cDNA library, Nagasaki et al. (1999) isolated a cDNA encoding BCAS2, which they termed DAM1 (DNA amplified in mammary carcinoma-1). The deduced 225-amino acid protein contains a central potential N-glycosylation site and multiple N-terminal potential phosphorylation sites. Northern and Southern blot analyses revealed expression of a 1.5-kb transcript at various levels in multiple breast cancer cell lines, with greatest amplification in BT-20 and MCF-7 cells. BCAS2 was ubiquitously expressed in normal tissues. Neubauer et al. (1998) reported the presence of a sequence identical to BCAS2 in a putative spliceosome-associated protein, which they termed SPF27.
