CLEC2 protein has a single carbohydrate recognition domain (CRD) with 6 conserved cysteine residues. Like CD94 and OLR1, the CLEC2 CRD lacks the amino acids necessary for calcium-dependent carbohydrate binding. CLEC2 also possesses a cytoplasmic tyrosine-based motif (DxYxxL) reminiscent of the motif essential for efficient endocytosis and basolateral expression of the asialoglycoprotein receptor (ASGR1), and it has several potential phosphorylation sites. Northern blot analysis revealed selective expression of a 1.0-kb transcript in liver, while RT-PCR analysis detected expression in myeloid and NK cells but not in other lymphocytes. Flow cytometric analysis demonstrated surface expression in CLEC2-transfected cells. Immunoprecipitation analysis indicated expression of a 33-kD protein.
