Comparison of the sequence of the human NOT3 protein to that of the 836-amino acid yeast NOT3 protein revealed 24% overall identity, with 41% identity in the first 232 amino acids. Compared to human NOT3, yeast NOT3 has a C-terminal extension. The 560-amino acid yeast NOT5 protein is highly similar to yeast NOT3 in its N-terminal region.
The human NOT3 protein shares 24% overall sequence identity with yeast NOT5, with 39% identity in the N-terminal 212 residues. The C-terminal domain of human NOT3 is extremely rich in proline and serine residues. Both recombinant human NOT3 protein expressed in mammalian cells and endogenous NOT3 from HeLa cells migrated as 120-kD proteins in SDS-polyacrylamide gels.
