CALHM2, deduced 323-amino acid protein contains 4 hydrophobic regions and shares 26% identity with CALHM1. Bioinformatic analysis suggested that CALHM2 is widely expressed.By genomic sequence analysis, Dreses-Werringloer et al. (2008) mapped the CALHM2 gene to chromosome 10q24.33, near the CALHM1 gene.CALHM1 decreased total extracellular accumulation of A-beta peptides, including the pathogenic peptides A-beta-40 and A-beta-42, in APP-expressing mouse neuroblastoma cells that had been exposed to Ca(2+) depletion followed by Ca(2+) add back. Inhibition of endogenous CALHM1 expression by RNA interference led to significantly increased A-beta levels in differentiated human neuroblastoma cells after Ca(2+) add back.
