Cell surface carbohydrates modulate a variety of cellular functions and are typically synthesized in a stepwise manner. HNK1ST plays a role in the biosynthesis of HNK1, a neuronally expressed carbohydrate that contains a sulfoglucuronyl residue.
Sequence analysis predicted that the 356-amino acid type II transmembrane protein contains 3 potential N-glycosylation sites and a conserved RDP sequence that is found in other Golgi-associated sulfotransferases. Northern blot analysis revealed wide expression of a 3.0-kb HNK1ST transcript that was most prominent in adult and fetal brain and adult testis and ovary.Functional analysis indicated that HNK1ST transfers a sulfate group to the appropriate substrate.
