Carbohydrate Sulfotransferase 12 (CHST12)
Chondroitin 4-sulfotransferases, such as CHST12, add sulfate to position 4 of N-acetylgalactosamine residues in chondroitin. The deduced 414-amino acid protein has a calculated molecular mass of 48.3 kD. C4ST2 has an N-terminal transmembrane domain and 4 regions of homology with other sulfotransferases: a 5-prime-phosphosulfate-binding site, a 3-prime-phosphate-binding site, and 2 alpha-helical domains near the C terminus.
C4ST2 expressed by transfected Chinese hamster ovary cells increased incorporation of radiolabeled sulfate into chondroitin, chondroitin 4-O-sulfate, chondroitin 6-O-sulfate, and desulfated dermatan sulfate. C4ST2 did not use a substrate sulfated by HNK1ST or other acceptor substrates examined.
