Sodium butyrate (NaB) induces growth inhibition and apoptosis in the human pancreatic cancer cell line AsPC-1. To identify genes differentially regulated by NaB, Zhang et al. (1998) generated subtracted AsPC-1 EST libraries enriched for up- or downregulated transcripts using the suppression subtraction hybridization (SSH) technique. The authors assembled a 2.45-kb cDNA sequence with an open reading frame encoding a putative 734-amino acid protein. They confirmed the calculated molecular mass of 82 kD by in vitro transcription and translation. The putative protein sequence, which shows extensive homology to alpha-catenin, has potential casein kinase and protein kinase C phosphorylation sites and a tyrosine kinase phosphorylation site.
