Cai et al. (2003) demonstrated that epitope-tagged rat Mrgbp associated with the TRRAP/TIP60 (HTATIP) histone acetyltransferase complex in transfected HeLa cells. Within this complex, Mrgbp bound directly to MRG15 (MORF4L1) and MRGX (MORF4L2).The International Radiation Hybrid Mapping Consortium mapped the C20ORF20 gene to chromosome 20. A null mutation of Trrap in mice results in preimplantation lethality due to a blocked proliferation of blastocysts. Trrap-deficient cells failed to sustain mitotic arrest despite chromosome missegregation and disrupted spindles, and displayed compromised cdk1 activity. Trrap is, therefore, essential for early development and required for the mitotic checkpoint and normal cell cycle progression.
