Bub1 Related Kinase (BRK1)

The C3ORF10 gene encodes a 9-kD protein involved in the complex process of actin nucleation.Eden et al. (2002) purified a multiprotein complex from bovine brain containing orthologs of human WAVE1 (WASF1), PIR121, NAP125, and HSPC300. Whereas recombinant WAVE1 is constitutively active, the WAVE1 complex is inactive. Actin polymerization studies suggested that RAC1 and NCK cause dissociation of the WAVE1 complex, which releases active WAVE1-HSPC300 and leads to actin nucleation.

Gautreau et al. (2004) purified WAVE2 from HeLa cells and determined that it associated in a complex with several other proteins, including HSPC300. HSPC300 showed direct interaction with the WAVE2 protein.
