Using suppressive subtractive hybridization, Singh et al. (2005) cloned a novel androgen repressible gene, which they called Admp (androgen downregulated gene expressed in mouse prostate), from the hypogonadal (hpg) mouse prostate. By database searching with the mouse sequence as query, Singh et al. (2005) identified the human homolog, C3ORF57, which encodes a deduced 76-amino acid protein that shares 85.5% sequence identity with mouse Admp. In situ hybridization with a DIG-labeled C3ORF57 antisense probe showed a signal in the epithelium of the human prostate. C3ORF57 expression in the prostate cancer cell line LNCaP was downregulated in the presence of androgens.Singh et al. (2005) determined that the C3ORF57 gene contains 3 exons and spans 70 to 90 kb. The ORF is contained entirely within exon 3.
