The predicted 1,327-amino acid protein is 48% identical to the quail neuronal cell cycle withdrawal protein Qn1. KIAA1009 is a coiled-coil protein containing a AAA ATPase domain, a Walker A motif, and domains involved in microtubule stabilization. Western blot analysis of rat pheochromocytoma PC12 cells detected a major 150-kD protein and minor 90- and 300-kD proteins. Cell fractionation showed that the 150-kD form was cytoplasmic, the 90-kD form was nuclear, and the 300-kD form was present in the cytoskeletal fraction. Confocal microscopy localized KIAA1009 at the spindle pole with alpha-tubulin (TUBA) and at the centrosome with gamma-tubulin (TUBG). Immunoprecipitation analysis revealed that the 150-kD KIAA1009 form was associated with TUBA at microtubules.
