CNKSR1 is a necessary element in receptor tyrosine kinase pathways, possibly as a tyrosine phosphorylation target. CNK1 physically associated with rhophilin, another Rho effector, and with RALGDS, a Ras effector, suggesting that CNK1 acts as a scaffold protein and mediates cross talk between these 2 GTPase signaling pathways.Cnk encodes a protein containing several protein-protein interaction domains, including SAM (sterile alpha motif), PDZ, and PH (pleckstrin homology) domains, suggesting that it brings different signaling molecules together. CNK is required in multiple receptor tyrosine kinase pathways where it appears to be a tyrosine phosphorylation target. CNK physically interacts with RAF and appears to localize to cell-cell contact regions.
