Connector Enhancer Of Kinase Suppressor Of Ras 2 (CNKSR2)
CNKSR2 encodes a multidomain protein that functions as a scaffold protein to mediate the mitogen-activated protein kinase pathways downstream from Ras. This gene product is induced by vitamin D and inhibits apoptosis in certain cancer cells.
The full-length CNK2A protein contains 1,034 amino acids, whereas the C-terminally truncated CNK2B protein contains 899 amino acids. Like Drosophila Cnk, both CNK2 isoforms contain SAM, CRIC, and PDZ domains in their N-terminal halves and a PH domain in their C-terminal halves. CNK2A also has a C-terminal PDZ binding motif that is absent in CNK2B. Immunofluorescence analysis localized both CNK2A and CNK2B to the lateral membrane, but not the apical portion, of transfected canine kidney cells.
