DEP Domain Containing Protein 1 (DEPDC1)
DEPDC1 plays a role in bladder cancer cell growth.The deduced DEPDC1-V1 and DEPDC1-V2 proteins contain 811 and 527 amino acids, respectively. Both isoforms contain an N-terminal DEP (Dishevelled/EGL-10, pleckstrin) domain. RT-PCR analysis detected increased DEPDC1 expression in 17 of 21 bladder cancers. Northern blot analysis of normal tissues detected a 5.3-kb transcript in testis, and 2 transcripts of 5.3 and 4.6 kb were detected in 4 bladder cancer cell lines. Immunohistochemical studies detected DEPDC1 in bladder cancer cells, but no staining was detected in normal bladder. Western blot analysis of bladder cancer cell lines detected 93.1-kD and 61.5-kD bands corresponding to the 2 isoforms. Both proteins localized to the nucleus in bladder cancer cells.
