DACT1 contains 4 coding exons spanning about 6.8 kb.The deduced 836-amino acid protein shares 60% amino acid identity with Xenopus Dapper, including complete identity in the last 28 amino acids of the C terminus, which include a consensus PDZ-binding motif.The deduced 799-amino acid protein contains 7 Dapper homology domains, including a leucine zipper, a serine-rich region, and a PDZ-binding domain.
DACT1 shares 28.8% amino acid identity with DACT2. In silico expression analysis indicated that DACT1 was expressed in a wide range of normal and tumor tissues.Dapper inhibited activation of JNK by DSH. Depletion of maternal Dapper RNA from Xenopus embryos resulted in loss of notochord and head structures.
