The folding pattern of the death domain shows that it belongs to what is known as death domain fold, an arrangement also observed in other protein modules. Proteins containing a death domain are members of a larger protein family called death domain superfamily.The death domain (DD) is a homotypic protein interaction module composed of a bundle of six alpha-helices. DD is related in sequence and structure to the death effector domain (DED) and the caspase recruitment domain (CARD), which work in similar pathways and show similar interaction properties. DD bind each other forming oligomers. Mammals have numerous and diverse DD-containing proteins.
