DHRS3 has a calculated molecular mass of 33.5 kD. It has all the characteristics of SDRs, including a cofactor-binding Rossman-fold domain and highly conserved motifs in the nucleotide-binding site and active site. Northern blot analysis detected a 1.8-kb RETSDR1 transcript in placenta, lung, liver, kidney, pancreas, and retina, but not in brain. A transcript of about 1.4 kb was detected in heart and skeletal muscle. Immunohistochemical analysis of bovine retina showed strong Retsdr1 staining of the cone outer segments, but not the rod outer segments, with diffuse labeling of the remainder of the cone photoreceptors. All cone types were immunoreactive, as were a subset of somata localized in the inner nuclear layer. Analysis of monkey and human retina sections produced similar results.
