DnaJ/HSP40 Homolog Subfamily C, Member 24 (DNAJC24)
Diphthamide is a unique posttranslationally modified histidine found only in translation elongation factor-2 (EEF2). This modification is conserved from archaebacteria to humans and serves as the target for ADP-ribosylation and inactivation of EEF2 by diphtheria toxin (DT) and Pseudomonas exotoxin A. DPH4 is 1 of several enzymes involved in synthesis of diphthamide in EEF2.
Liu et al. (2004) identified 2 variants of mouse Dph4 that encode proteins of 193 and 148 amino acids. Both isoforms contain an N-terminal DnaJ domain and a C-terminal CSL zinc finger domain. Mouse and human DPH4 share 83% amino acid identity.The International Radiation Hybrid Mapping Consortium mapped the DPH4 gene to chromosome 11.
