DNAJC3 contains multiple tetratricopeptide repeat (TPR) motifs as well as the highly conserved J domain found in DNAJ chaperone family members. It is a member of the tetratricopeptide repeat family of proteins and acts as an inhibitor of the interferon-induced, dsRNA-activated protein kinase (PKR).The 58-kD inhibitor of the interferon-induced double-stranded RNA-activated protein kinase (PKR) is a cellular protein that is recruited by influenza virus to downregulate PKR, which in turn plays a major role in mediating the interferon response to viral infection.The inhibitor protein, also called p58, may also play a role in gene expression in uninfected cells and its overexpression in mouse 3T3 cells results in a transformed phenotype which rapidly forms tumors in mice.
