The cytoplasmic domain contains 7 putative phosphorylation sites and 6 N-myristoylation sites. The domain structures of DSCAML1 and DSCAM are identical, and they share 64% and 45% amino acid identity in their extracellular and cytoplasmic domains, respectively. In situ hybridization revealed Dscaml1 in the Purkinje cell layer of mouse cerebellum, in granule cells of the dentate gyrus and in pyramidal cells of the CA1 and CA3 regions of the hippocampus, and in olfactory bulb, with moderate expression in cerebral cortex and thalamus. Immunohistochemical analysis of transfected mouse fibroblasts and rat PC12 pheochromocytoma cells showed expression on the cell surface. In differentiated transfected PC12 cells, human DSCAML1 associated with axons, but not with dendrites.
