Ectopic expression of E2F7 in U2OS cells suppressed E2F target genes and led to an accumulation of cells in G1. E2F7 associated with E2F-regulated promoters in vivo, and this association increased in S phase. Chromatin immunoprecipitation (ChIP) assays showed that E2F7 bound the promoters of a subset of E2F-dependent genes, including E2F1 and CDC6 (CDC18L), in U2OS cells and human fibroblasts. Inhibition of E2F7 expression resulted in specific derepression of the E2F1 and CDC6 promoters.E2F7A lacks exon 12 and encodes a 728-amino acid protein, whereas E2F7B includes exon 12 and encodes a 911-amino acid protein. The E2F7A and E2F7B proteins differ only after amino acid 713, and both have 2 N-terminal E2F-type DNA-binding domains containing highly conserved residues involved in binding.
