E2F8 contains 2 N-terminal DNA-binding domains and shares 82.2% and 31.9% amino acid identity with mouse E2f8 and human E2F7, respectively. Real-time quantitative PCR detected abundant E2F8 expression in transformed cells lines of carcinoma and sarcoma origin. Western blot analysis of transfected HeLa cells revealed a major E2F8 protein of about 100 kD. Epitope-tagged E2f8 bound specifically to an E2F probe, and mutation analysis showed that residues within both DNA-binding domains that are involved in E2f8 dimerization were required for DNA binding. RT-PCR showed mild induction of E2f8 expression during S phase of mouse embryonic fibroblasts, and overexpression of E2f8 in mouse embryonic fibroblasts blocked proliferation
