FYVE, RhoGEF And PH Domain Containing protein 2 (FGD2)
FGD1 , the gene that is mutated in faciogenital dysplasia, encodes a GEF that specifically activates the Rho GTPase Cdc42. Rho GTPases are Ras superfamily members thought to be involved in cytoskeletal restructuring.Fgd2 and FGD1 share a high degree of sequence similarity that spans over 560 contiguous amino acids. Like FGD1, Fgd2 contains adjacent RhoGEF and pleckstrin homology (PH) domains, a second carboxy-terminal PH domain, and a distinctive FYVE domain. Northern blot analysis of mouse RNA showed that 3.0- and 4.2-kb Fgd2 transcripts are expressed in adult spleen and lung tissues, and that several prominent transcripts are expressed during mouse embryogenesis, suggesting that Fgd2 may be involved in embryonic development.
