EEIG1 in MCF-7 cells exposed to E2 compared to controls, similar to results observed in microarray experiments. Induction of gene expression was detectable at 0.001 nM of E2, with the highest levels of expression at 10 nM of E2. EEIG1 expression was observed in MCF-7 cells exposed to natural, synthetic, and xenoestrogens but was less evident with phytoestrogens or pesticides. E2-induced expression of EEIG1 was blocked by tamoxifen and ICI-182780 but not by cycloheximide and puromycin, suggesting that EEIG1 is directly regulated by E2. When 3 ER-positive and 3 ER-negative breast cancer cell lines were exposed to E2, EEIG1 expression was selectively induced in ER-positive cell lines, indicating that EEIG1 expression is dependent on the presence of a functional estrogen receptor.
