Using a proteomics approach to identify genes upregulated in breast cancer cell membranes, followed by database analysis and PCR of a pooled testis, fetal lung, and B-cell cDNA library, Adam et al. (2003) cloned BCMP101. The deduced protein contains 310 amino acids. RT-PCR and immunohistochemical analyses demonstrated low BCMP101 expression in multiple normal tissues. However, high levels of BCMP101 mRNA were detected in breast carcinoma cells, with expression upregulated more than 2-fold in 6 of 7 breast carcinomas tested compared with adjacent normal tissue. Fluorescence-tagged BCMP101 showed widespread intracellular localization and significant expression on the plasma membrane, particularly in areas of cell-cell contact.
