Chang et al. (2000) cloned P29 by a yeast 2-hybrid screen of a HeLa cell cDNA library with GCIP (CCNDBP1) as bait. The deduced 243-amino acid protein has a predicted molecular mass of 29 kD. It contains 2 bipartite nuclear localization signals, another basic region of nuclear localization signal RKKKRK, and 2 potential polyadenylation signals in the 3-prime untranslated region.
Northern blot analysis detected expression of 1.35- and 0.9-kb transcripts in all tissues examined, with highest expression in heart, skeletal muscle, and kidney. In vitro translation followed by Western blot analysis revealed a single major protein with an apparent molecular mass of 29 kD. Fluorescence-tagged P29 localized to the nucleus of transfected HeLa cells.
