The predicted 370-amino acid SREB2 protein contains several features of the GPCR family, including 7 transmembrane domains, potential N-linked glycosylation sites in the N-terminal extracellular domain, and conserved cysteine residues, which may form a disulfide bridge, in the first and second extracellular loops.
However, SREB2 shares additional characteristics with SREB1 and SREB3 that are not also shared with other GPCRs, including identical amino acid substitutions of residues conserved in other GPCRs, an absence of prolines in the fifth transmembrane domain, and a relatively large conserved third cytoplasmic loop that has no sequence similarity to the third cytoplasmic loops of other GPCRs. Human SREB2 and rat Sreb2 have identical amino acid sequences.
