G Protein Coupled Receptor, Family C, Group 5, Member D (GPRC5D)
GPRC5D shares significant amino acid identity with other GPRC5 members, including RAIG1 (42%), GPRC5B (27%), and GPRC5C (33%). The human and mouse proteins share 83% identity over the first 298 amino acids. However, mouse Gprc5d lacks the 45 C-terminal amino acids found in the human protein, which are encoded by exon 2. Northern blot and RT-PCR analyses revealed high expression of GPRC5D in pancreas; medium expression in kidney, small intestine, spleen, and testis; low expression in lung, colon, leukocytes, prostate, and thymus; and no detectable expression in brain, heart, liver, placenta, skeletal muscle, and ovary. Fluorescence-tagged GPRC5D localized to the cell membrane of transfected HEK293 cells and showed microclusters of higher expression.
