GC Binding Factor (GCF)
GCF represses transcription of certain genes and is encoded by a 3-kb mRNA.the 5-prime end is 308 nucleotides shorter than the original sequence, that a 'T' at position 787 is not observed, and that there is an insertion of 114 nucleotides between positions 851 and 852 of the previous sequence. The revisions resulted in a cDNA consisting of 2,661 nucleotides encoding a 781-amino acid protein. SDS-PAGE analysis determined that GCF is expressed as an approximately 120-kD protein. Gel shift analysis also revealed that the revised GCF has no sequence-specific DNA-binding activity. Takimoto et al. (1999) concluded that to avoid errors in the sequencing it is important to confirm cDNA sequences by isolating and sequencing more than 1 clone and by comparing the cDNA with the genomic DNA sequence.
