GTFs assemble in a complex on the DNA promoter and recruit the RNA polymerase. GTF3C family proteins are essential for RNA polymerase III to make a number of small nuclear and cytoplasmic RNAs, including 5S RNA, tRNA, and adenovirus-associated (VA) RNA of both cellular and viral origin.
Sequence analysis showed that the deduced 822-amino acid GTF3C4 protein contains 2 potential zinc finger motifs at the C terminus. Immunoblot analysis indicated that the expressed cDNA encodes an approximately 90 kD protein that copurifies with the remainder of the GTF3C complex and interacts with GTF3B in the recruitment of RNA polymerase III. Functional analyses showed that GTF3C4 has intrinsic histone acetyltransferase activity.
