NP60 contains 2 nuclear localization signals, an N-terminal PWWP domain and an AT-hook, and a C-terminal NAD-binding domain. Western blot, RT-PCR, and immunohistochemistry showed that NP60 localizes to the nucleus in a variety of cell lines, with robust expression in mouse macrophage RAW274.7 cells.
Cotransfection experiments showed that NP60 specifically induces phosphorylation of p38-alpha in response to sorbitol-induced stress, but not JNK1 or ERK2, and that the AT-hook and NAD-binding domains of NP60 are required for this phosphorylation. A series of phosphorylation assays determined that NP60 does not directly act on p38-alpha, but instead enhances activity of the upstream kinases MAP2K4 and MAP2K6.
