The deduced protein consists entirely of 7 kelch domains, which fold into beta-propeller structures capable of participating in protein-protein interactions. Database analysis suggested that KLHDC8B is widely expressed. Immunofluorescence analysis of HeLa cells showed that KLHDC8B concentrated in the midbody of the cytoplasmic bridge linking mitotic daughter cells just prior to cytokinesis. Flow cytometry, Western blot analysis, and RT-PCR of synchronized HeLa cells demonstrated that KLHDC8B was transcribed predominantly during S phase, translated exclusively during mitosis and cytokinesis, and rapidly degraded after cell division.depletion of KLHDC8B expression in HeLa cells via RNA interference resulted in an increase in the frequency of binucleated cells, with an increased proportion of cells with 4N DNA content.
