Keratin 74 (KRT74)
KRT74 belongs to the family of type II keratins that are specifically expressed in the inner root sheath of hair follicles. The deduced 529-amino acid protein has a calculated molecular mass of 57.9 kD. K6IRS4 shares a highly conserved rod domain and flanking non-alpha-helical H1 and H2 subdomains with K6IRS1, K6IRS2 , and K6IRS3. The head and tail domains are less related. Western blot analysis detected expression in root sheath extracts, but not in foot sole extracts. In situ hybridization and indirect immunofluorescence showed expression of K6IRS4 restricted to the Huxley layer of the inner root sheath. K6IRS4 was expressed in specialized Huxley cells, termed 'Flugelzellen,' which means 'winged cells,' along the area of differentiated Henle cells. K6IRS4 staining of the foot processes of Flugelzellen overlapped staining for K6IRS1.
