The deduced 636-amino acid mouse protein contains a predicted signal peptide, a leucine-rich N-terminal domain, 6 leucine-rich repeat domains, a leucine-rich C-terminal domain, Ig C2-like domain, fibronectin type III domain, transmembrane region, and an ESTV PDZ-binding motif. The LRFN family members share similarity with proteins of the Slit family , the Amigo family (AMIGO1), and the Nogo66 receptor (RTN4R).Northern blot analysis of adult mouse tissues detected Lrfn4 expression in brain and testis. In situ hybridization in mouse embryos demonstrated strong Lrfn4 expression in telencephalic and diencephalic vesicles at embryonic day (E) 11.5 to E17.5. Weak expression of Lrfn4 was detected from E4.5 to E9.5 with an increased expression around E10.5.
