By database analysis, Mi et al. (2004) identified LRRN6D, which they called LINGO4. LINGO4 belongs to a leucine-rich repeat protein family and shares 44% amino acid identity with LINGO1 (LLRN6A).Inhibitors of myelin prevent axonal regeneration in the adult CNS, and this inhibition involves modulation of RhoA (ARHA) activity by NGR1 (RTN4R) and NGFR. Mi et al. (2004) identified LINGO1 as an NGR1 binding partner and found that LINGO1 was a component of the NGR1/NGFR signaling complex. In nonneuronal cells, coexpression of human NGR1, NGFR, and LINGO1 conferred responsiveness to oligodendrocyte myelin glycoprotein (OMG), as measured by RhoA activation. A dominant-negative LINGO1 construct attenuated myelin inhibition in transfected primary rat neuronal cultures.
