Fad158 was among about 100 genes expressed in the early stage of adipocyte differentiation in the 3T3-L1 mouse fibroblast cell line. They found that Fad158 expression was quickly and transiently expressed in early differentiation. Overexpression of Fad158 in NIH-3T3 cells, a mouse fibroblast cell line that does not usually differentiate into adipocytes, in the presence of a ligand for PPAR-gamma resulted in differentiation; overexpression of Fad158 alone in these cells did not. Expression levels of PPAR-gamma and SREBP1 , and its target genes lipoprotein lipase and adipocyte fatty acid-binding protein-2, were elevated during differentiation, whereas expression levels of C/EBP family members were not, suggesting that FAD158's role in adipogenesis is linked with the PPAR-gamma, and not the C/EBP, pathway.
