Using a PDZ domain-binding peptide as bait in a yeast 2-hybrid screen of a mouse cerebellar cDNA library, Rice et al. (2001) cloned mouse Lnx2. The deduced 687-amino acid protein contains a RING finger domain, followed by a motif similar to a PTB-binding motif and 4 PDZ domains. Northern blot analysis detected Lnx2 expression in all mouse tissues examined. In situ hybridization of embryonic mouse detected Lnx2 expression in several tissues, although highest expression was in forebrain. Within adult brain, Lnx2 was expressed in several regions and nuclei in a pattern that often complemented that of Lnx1.mouse Lnx2 formed oligomers and heteromers with Lnx1, and they determined that Lnx2 has at least 2 separate oligomerization sites. Lnx2 bound Numb family proteins through its PTB-binding motif.
