Ly6/PLAUR Domain Containing Protein 1 (LYPD1)
The deduced 141-amino acid protein has a predicted molecular mass of approximately 44 kD, confirmed by Western blot analysis. LYPD1 contains an N-terminal signal sequence and a leukocyte antigen-6 (Ly6)/uPAR (PLAUR) domain, or LU domain, characteristic of glycosylphosphatidylinositol (GPI)-linked cell surface glycoproteins and similar to the LU domain of CD59. The LYPD1 LU domain contains 12 cysteine residues potentially involved in 6 intramolecular disulfide bonds.
Upregulation of p53 (TP53) played a role in the HeLaHF cell revertant phenotype; however, RNAi-mediated downregulation of LYPD1 did not inhibit anoikis (detachment-induced apoptosis) in HeLaHF cells, in contrast to the inhibition of anoikis detected upon RNAi-mediated downregulation of p53.
