Mammalian Map6d1 is a neuronal protein that binds calmodulin, displays microtubule binding and stabilizing activities, and localizes to Golgi material in neurites where it may play a role in synaptic function. The deduced 131-amino acid mouse protein shares significant homology with N-STOP over the N-terminal domain and a microtubule-stabilizing Mn module, both of which contain calmodulin-binding motifs. Western blot analysis detected mouse Sl21 exclusively in brain tissue with low levels in newborn brain on postnatal day 0 (P0) that increased after P10. Sl21 protein was detected in neurons in primary cultures but not in glial cells or neuronal cell lines. Immunohistochemistry of cultured differentiating hippocampal neurons localized Sl21 to the somatic Golgi apparatus, as well as to neurite vesicular Golgi in late stages of neuronal differentiation.
