Mannosidase Alpha Class 1A Member 2 (MAN1A2)
Alpha-mannosidases function at different stages of N-glycan maturation in mammalian cells. MAN1A2 gene contains at least 13 exons and spans at least 60 kb.The full-length cDNA predicts a 641-amino acid protein with characteristics of a type II membrane protein and a potential N-glycosylation site. The MAN1A2 protein is 94% identical to the homologous mouse protein. Northern blot analysis identified transcripts ranging from 3.5 to 9.5 kb, with highest levels of expression in placenta and testis. A truncated soluble form of alpha-1,2-mannosidase IB protein was expressed in the yeast strain Pichia pastoris, and alpha-1,2 mannosidase activity was detected in the growth medium. Western blot analysis detected a recombinant alpha-1,2-mannosidase protein of the expected size (67 kD).
